单B细胞抗体技术
虽然杂交瘤技术和噬菌体展示技术已经在单克隆抗体生产中广泛应用 , 但它们依然存在较难克服的缺点制约着抗体生产过程 。 近些年 , 为了克服杂交瘤技术细胞融合效率低 , 噬菌体展示技术导致重链、轻链的天然同源配对丢失等问题 , 单B细胞抗体技术正被逐步开发和应用 。
简单来说 , 单B细胞抗体技术包含以下几个步骤:
a.从外周血或免疫器官中初步提取淋巴细胞;
b.使用磁性活化细胞分选(MACS)或荧光活化细胞分选(FACS)技术鉴定和分离出特定的B细胞;
c.将分离出的B细胞进行单细胞培养;
d.使用逆转录聚合酶链式反应(RT-PCR)和抗体特异性引物鉴定单B细胞分泌抗体的特异性;
e.扩增特异抗体基因;
f.将抗体基因克隆到表达载体中 , 并在细菌或细胞系统中表达;
g.纯化表达产生的抗体 , 并用ELISA等方法进行评估 。
单B细胞抗体技术现在已广泛用于人和小鼠单克隆抗体生产 , 如一些治疗性中和单抗 , 可用于治疗多种疾病 , 包括癌症、自身免疫性疾病和传染性疾病亡[13,14] 。
